چکیده

Introduction Phosphatases are enzymes that remove phosphate groups from substrates. Phosphorylated compounds are widely distributed in living systems. They serve as storage forms for energy (e.g., ATP and phosphocreatine), as components of informational macromolecules (i.e., nucleotides and deoxynucleotides), as allosteric effectors of certain enzymes (e.g., fructose-1-6bisphosphate), and as second messengers (e.g., cAMP, cGMP, inositol phosphates). Phosphorylation-dephosphorylation reactions of proteins, mediated by protein kinases and protein phosphatases, modulate many enzyme activities (phosphorylase, pyruvate dehydrogenase, etc.). It is therefore not surprising that phosphatases of many kinds can be extracted from many tissues. In general, these enzymes catalyze the hydrolysis of phosphate monoesters as follows:

Studies on kinetic properties of acid phosphatase from nuclei-free rat liver homogenate using different substrates.

Kinetic properties of rat liver acid phosphatase were evaluated using the conventional synthetic substrates sodium beta glycerophosphate (betaGP) and p-nitrophenyl phosphate (PNPP) and physiologically occurring phosphate esters of carbohydrates, vitamins and nucleotides. The extent of hydrolysis varied depending on the substrates; phosphate esters of vitamins and carbohydrates were in general p...

Temperature Effect on Swelling Properties of Commercial Polyacrylic Acid Hydrogel Beads

Due to the fact that many application processes take place at different temperature, better examining and understanding of swelling properties as a function of temperature is essential for such application. The temperature effect on the swelling properties of commercial polyacrylic acid (PAA) hydrogel beads was investigated as a function of time and temperature. Gel bead size, swelling equilibr...

Influence of Subunit Interface Mutations on Kinetic and Dynamic Properties of Alkaline Phosphatase from E. coli

Mutations, replacing amino acids involved in the formation of hydrogen bonds between subunits of dimeric alkaline phosphatase, have been introduced. Influence of mutations on kinetic properties and structural stability of mutant enzymes was established. In addition, alterations in protein dynamic properties have been studied using room temperature phosphorescence. Kinetic properties of both mut...

The Kinetic and Thermal Degradation Studies of N-(4-Acetyl-phenyl)-acetimidic Acid Pyridine-3-yl ester Metal(II) Complexes

Schiff base complexes of transition metals are of particular interest to inorganic chemists because of their structural, spectral and chemical properties, which are often strongly dependant on the nature of the ligand structure. Large number of metal (II) complexes with Schiff-base ligands has been extensively studied for their interesting structural specialties, applications and properties. Th...

Biochemical characterization of the class B acid phosphatase (AphA) of Escherichia coli MG1655.

The AphA enzyme of Escherichia coli, a molecular class B periplasmic phosphatase that belongs to the DDDD superfamily of phosphohydrolases, was purified and subjected to biochemical characterization. Kinetic analysis with several substrates revealed that the enzyme essentially behaves as a broad-spectrum nucleotidase highly active on 3'- and 5'-mononucleotides and monodeoxynucleotides, but not ...
